Reading the sequence
Three features of Gly-Glu-Pro-Pro-Pro-Gly-Lys-Pro-Ala-Asp-Asp-Ala-Gly-Leu-Val determine how this material behaves at the bench.
The three consecutive prolines at positions 3-5, with a fourth at position 8, make roughly a quarter of the chain conformationally locked. Proline’s ring restricts backbone rotation, and rigid segments are poor substrates for endopeptidases, which require an extended geometry to bind.
No cysteine means no disulfide chemistry: no reduction risk, no dimerization, no scrambling. Reducing agents in a buffer are simply irrelevant here.
No methionine and no tryptophan removes the two most oxidation-prone residues. Light and dissolved oxygen matter far less for this peptide than for a sequence such as Semax.
The net result is an unusually forgiving research material. One lysine and a free N-terminal amine against one glutamate and two aspartates gives a roughly balanced, mildly acidic molecule with good aqueous solubility and — because there are only two basic sites — a comparatively low counterion load.
