Triple-receptor design

GIP, GLP-1 and glucagon belong to the same peptide hormone family, sharing an evolutionary origin and a common class B G-protein-coupled receptor. That homology is what makes multi-receptor engagement possible at all — but each receptor has distinct preferences at particular positions, so a sequence engaging all three must satisfy three overlapping and partly conflicting sets of constraints while retaining the modifications needed for stability.

Why no sequence is published here

A plain 39-letter residue string would omit the Aib substitutions that block dipeptidyl peptidase-4 cleavage, the acylation site, the linker chemistry and the fatty acid — that is, most of what distinguishes this molecule from a generic peptide of the same length. Identity confirmation rests on mass spectrometry against the complete modified structure, which is what the lot certificate records.

Analytical and handling notes

Strongly hydrophobic for its length, owing to the lipid: late reversed-phase retention, readily adsorbed to plastic, and aggregation-prone under agitation. Vortexing causes visible loss. At 4,731.4 g/mol a single-residue deletion shifts mass by 1-3% and a +16 oxidation by 0.3%, so high-resolution measurement or a well-resolved LC-MS gradient is the appropriate method.
